Rapid Transient Transcriptional Adaptation to Hypergravity in Jurkat T Cells Revealed by Comparative Analysis of Microarray and RNA-Seq Data

Cellular responses to micro- and hypergravity are rapid complex appear within the first few seconds of exposure. Transcriptomic analyses a valuable tool analyze these genome-wide cellular alterations. For better understanding dynamics upon altered gravity exposure, it is important compare different time points. However, since most experiments designed as endpoint measurements, combination cross-experiment meta-studies inevitable. Microarray RNA-Seq two main methods study transcriptomics. In field research, both frequently used. generation data sets difficult time-consuming therefore number available in this research limited. study, we investigated comparability microarray applied results comparison transcriptomics between conditions during real flight platforms centrifuge experiment identify temporal adaptation processes. We performed comparative on an Affymetrix HTA2.0 paired-end set originating from same Jurkat T cell RNA samples short-term experiment. The overall agreeability was high, with sensitivity analysis. showed weaknesses level single upregulated genes, likely due its normalization approach. On aggregated biotypes, chromosomal distribution, gene sets, technologies equally well. performance detection gravity-related splicing events. found that all initially transcripts fully adapted after 15 min concluded expression response transient reversible. Based combined multiple-platform meta-analysis, could demonstrate transcriptional hypergravity, differential ATPase subunits ATP6V1A ATP6V1D, cluster differentiation (CD) molecules CD1E, CD2AP, CD46, CD47, CD53, CD69, CD96, CD164, CD226 hypergravity. experimentally possible develop methodological evidence for meta-analysis individual data.
